(Senior) Scientist, TCR screening & Validation, TCR Selection Facility

Neogene Therapeutics is a pre-clinical stage biotechnology company aiming to change the paradigm of treatment for solid cancers. Our mission is to provide cancer patients with T cell therapies targeting mutated proteins called neo-antigens, which are present on cancer cells as a consequence of DNA mutations. Neogene is developing novel technologies to enable the engineering of ‘designer T cells’ with neo-antigen specificities that display an optimized ability to seek and destroy cancer cells.

We are seeking a highly motivated individual to join our personalized TCR discovery team in Amsterdam as a (Senior) Scientist, TCR screening and validation. The personalized TCR discovery team is responsible for isolating neo-antigen specific TCRs from a cancer patient’s tumor biopsy & characterizing these tumor-derived TCRs. Following isolation and characterization, neo-antigen specific TCRs are selected for the clinical manufacture of a fully personalized multi-specificity neo-antigen specific TCR engineered T cell therapy product that is used to treat the cancer patient.  Reporting to the Associate Director of the personalized TCR discovery team at the newly established TCR selection facility in Amsterdam, the individual will assume responsibility for supervising a team of research associates performing experiments to isolate and characterize patient-derived neo-antigen specific TCRs for clinical use.

Responsibilities include, but are not limited to:

  • Supervise and hands-on support of laboratory activities related to the TCR screening & validation process steps in the TCR selection facility. The facility runs under a quality system that is based on the principles of Good Laboratory Practice (GLP).
  • Supervise a team of research associates performing experiments to isolate and characterize patient-derived neo-antigen specific TCRs in a manner compliant with standards set out by relevant accredited agencies.
  • Provide day-to day status updates of ongoing laboratory activities related to clinical across an integrated cross-functional team to ensure collaboration
  • Prepare data reports and presentations to inform the selection of personalized TCRs for clinical use.
  • Serve as a subject matter expert for methods utilized for TCR discovery & collaborate with the Neogene research team to support the identification, validation and implementation of process step improvements.
  • Drafting and reviewing of protocols, SOPs, batch records, technical reports and regulatory documents to support clinical development.
  • Mentor and develop more junior staff
  • Other duties as assigned.

Requirements:

  • PhD with at least 3 years of relevant work experience in Immunology or a related field. Experience of or an affinity for working in a GLP or GMP environment. Industry experience is a plus. Candidates with relevant experience may be eligible for a position as a Principal Scientist (at the company’s discretion).
  • Deep knowledge of immunological techniques, including magnetic bead-based cell separation of human immune cells (T cells, B cells, Dendritic cells and others), phenotypic and functional evaluation of immune cells by flow cytometry (surface and intracellular staining, proliferation tracking, cytotoxicity assays) and genetic modification of primary immune cells and cell lines utilizing retroviruses and gene editing tools (e.g. CRISPR-Cas9).
  • Preference for candidate with experience of drafting and reviewing documentation to support clinical development
  • Preference for candidates with experience of working within a formal quality management system.
  • Experience of working in a cGMP or ISO 13485 production environment is a plus
  • Excellent organizational and managerial skills with experience in the supervision of junior research staff
  • Comfortable with electronic document management systems and authoring tools
  • A strong sense of urgency with regards to work duties and the ability to deliver results based on challenging timelines.
  • Strong verbal and written communication skills and the ability to communicate results to a diverse cross-functional audience.
  • Comfortable in a fast-paced growing company environment and able to adjust workload based upon changing priorities.
